Absolutely - I'd even go as far as saying that xtrabackup should be the first choice for setting up new slaves as it makes it so ridiculously easy. Because xtrabackup doesn't block at all, you don't have to stop your slave, so you won't accumulate lag. The other big thing about xtrabackup is that its backups can be quite slow, but its restores are really fast (only slightly longer than copying the files). mysqldump is usually the reverse, which is not what you want when you're in a mad panic trying to restore a backup!
